Penn's recreational drug use led to him criticizing Wyclef's presidential bid.
Penn told Larry King last month that he worried the singer’s campaign was simply "about a vision of flying around the world, talking to people."
He later penned a column for the Huffington Post criticizing the singer for being absent after the January earthquake in Haiti.
"None among us felt or expressed anger toward it, but rather a universal sadness for his silence, as he is America's most admired cultural link to Haiti," the actor wrote.
Jean lashed out at Penn’s diatribe while performing his 2004 hit "President" at Hot 97's On the Reggae Trip last week.
"I got a message for Sean Penn: Maybe he ain't see me in Haiti because he was too busy sniffing cocaine," he sang.
Penn's rep shot back at Jean's comments, calling them "highly irresponsible and false."
"Mr. Jean is clearly unfamiliar with the physical demands put upon volunteers in Haiti. As aid workers there, the notion of depleting the body’s immune system thru the use of illicit drugs is ludicrous. More specifically, J/P Haitian Relief Organization (a.k.a. JPHRO) has a ZERO tolerance policy for any and all illegal drugs," said Mara Buxbaum. "As the leader of this organization, Sean Penn has not only set this policy, but adheres to it. That Mr. Jean would make such a false accusation is reckless and saddening, but not surprising."
The singer also took issue with Pras Michel, with whom he used to perform in The Fugees, after the former criticized his strategy for revitalizing Haiti.
In response, Jean said on Friday, "I got a message for Praswell, even though you don't want to support me, I got love for you, even though you only kicked 8 bars in the Fugees."
Weeks after he announced his plan to run for President of Haiti at the beginning of August, the country's electoral officials declared Jean was ineligible to run. Though they did not give a reason for the disqualification, reports suggested it was because the singer had not lived in Haiti for five years, a requirement for candidacy.
GV Says: Wow...way out of line Wyclef, way out of line. And that my friend is a great example of why your bid got denied. As mad as you are with Sean Penn, to publicly suggest drug use is reckless...and the jab he made at Pras was unnecessary. Be a bigger man...SMH at that childishness.
